Considering the -- to my knowledge -- relative numbers of biological children vs. adopted children, when two fictional characters have a parent-child relationship I assume biological unless there is reason to believe otherwise.

On the other hand, if one character is known to have parented another character, their referring to that character as their child does not provide any additional evidence of biological relationship.

I don't know the fandom or the argument, OP, so I have no idea whether I agree with you.

In the early part of the comic, you're led to believe that she's adopted. There are hints later on that she's not, which sparked debate in fandom, and during that, people would say "well, he calls her his daughter, so they must be biologically related."
