It's because of the endless jokes about Dean's sexuality on the show - hiding an erection when he sees Castiel fixed up, having a cowboy fetish, crushing on Dr Sexy, having his siren turn into a man etc. etc.

They hardly ever do jokes like that about Sam.

Plus there's the fact that Dean used to be visibly uncomfortable around gay men which could be interpreted as denial or repression.
