Off the top of my head...

- If you're replying to a job posting, include action words from it in your cover letter and resume. If not, link your resume and cover letter by using the same action words.

- Make sure you address it to someone specific. Find out their name and include it. Don't use a general salutation.

- Tell them why the company would benefit hiring you and what you can bring to the table.

- Prove that you're interested in the company and that you have knowledge about them.

- Thank them for their time and say something about hoping to meet them.
