Ned Stark: a major POV character in the first book, the one that directly links to every other POV in AGOT except Dany (and even then, she's an exile because of the Rebellion, and Jorah is Northern lord with a grudge against Ned). In the show, he's the main character.

Robert, Robb, and Joffrey: all Kings (also Viserys, Renly, and Balon, but I don't consider them major characters)

Catelyn Stark: major POV character for the first 3 books/3 seasons. Comes back as a reanimated corpse (no longer POV) in books 4 and 5.

Tywin Lannister: the power behind the throne, dies at the end of book 3/season 4.

Unconfirmed deaths from ADWD (book 5)-
Jon Snow: may have died in a mutiny at the Wall. Jon, Dany, and Tyrion get the most focus/screentime and can be considered the "main characters". So if Jon is dead, he's the most important character to die since Ned. I doubt Jon is dead, but if he is, there's a good chance he'll be back.

Stannis Baratheon: last remaining King in the War of the 5 Kings. May have been killed by the Boltons, but maybe not.

Theon Greyjoy: POV character in books 2 and 5, last seen in Stannis' hands, probably won't make it unless Stannis dies.

And Jon Connington (POV book 5) is slowly dying of a horrible disease because he's an idiot who would have had a better chance if he cut off his hand.
