They literally explicitly say that they're not brothers and their relationship isn't brotherly in the strips, so no they're not.

(You can certainly see it as brotherly if you want, but it's not canon.)

That's not true. In the episode where little America is discovered, all the older European nations get in an argument over who gets to be his big brother. They may not be blood related, but the relationship is supposed to be that of an older brother and younger brother. Although, with the differences in parental and sibling behavior between Japan and the West, a lot of Western fans tend to read it as father/son ish.

Actually England outright tells him not to call him brother (don't remember if they cut that in the anime) once he actually meets him (the aforementioned conversations happened before). But even if you consider them in a brotherly relationship at the time, it's canon that they don't consider themselves that once they reach adulthood.

They do explicitly say they are not siblings and their relationship is not that of siblings. It's in volume... five I think? One of the African Front strips from Comic Birz.

And of course, in the Cleaning Out the Storage strip America also says it.

So no, they aren't.
