It's only easy to interpret Rui as trans if you have an incredibly shallow, black-and-white view of gender.

Wearing pretty clothes doesn't mean jack shit except he likes wearing pretty clothes. It'd be a different story if he showed even the slightest hint of dysphoria, or discomfort about his biological sex, or demonstrated any kind of desire to be female or be treated as female. But he doesn't. At all.

If you want trans headcanons, what about OD? Or, hell, you could even make a better argument for Hajime being trans because she uses masculine pronouns. It's very (very) flimsy, yes, but at least it relates to her behaviour rather than her appearance.
