Definitely start with the first book, "The Eyre Affair" (love of "Jane Eyre" is helpful but not required). It's not a series that you can do out of order - even though she's a government agent, and you'd expect it to be more 'monster of the week', the world builds on itself with each novel. Her relationships with her family also actually change from book to book, unlike a lot of detective-type novels, so that'd get confusing out of order.

There's also a spinoff series (well, there's only 2 so far) called the Nursery Crimes series. I find them very charming too, and they can be read on their own, but you'll get a lot more of the references/in-jokes if you wait until the point they branch off of the first series.
