I don't know what they were saying but a lot of people who didn't play the game either think they are siblings or that there is an age difference... both are not true. They are just two androids made by YoRHa. The most recent batch of androids, so probably just a few years old probably. Every character in the game is an android or machine, but YoRHa are like elite military androids that fight against the machines.

The boy (9S) is a scanner type android and the girl (2B) is an executioner type android. Every time the boy comes close to confidential information she has to kill him... but not really kill him... more like reset his memories. So she remembers everything but he doesn't.

The game doesn't really go in depth with the past of their relationship but according to side material they are the most important people in each others short existence. So her to be ordered to reset him so many times is painful for her. Later on 2B catches a fatal virus and ask another android called A2 to mercy kill her. 9S sees this happening and goes completely insane suicide revenge mission against A2.

What is this and what is the actual relationship?

It's NieR: Automata, and the relationship is that they're both androids who are soldiers fighting machines on Earth. (Yep, androids vs machines, the philosophical debate about if there's even a distinction between that does get mentioned in the story, but that's neither here nor there.) The one who's carrying the other is 2B, while the one being carried is 9S. 2B has a hardened, business-oriented, and distant attitude, but 9S is geeky, chipper, and honestly really likes 2B.

2B's attempts to keep herself emotionally distant away from 9S seems to be because 9S tends to get killed a lot (in very emotional fashion, as dying tends to be), but that's made difficult since everyone is an android, and 9S gets brought back to work with her again missing his memories, still chipper, dying horribly each time, time and time again. This sort of existence goes on for centuries.

Centuries? I didn't know they existed that long. I know they kill off YoRHa androids in batches to bring in the new ones.

I don't think it was centuries for YoRHa units specifically. The Pearl Harbor Descent was in 11941 AD, which was only 4 years before Nier: Automata itself, and that was A2's first mission to the surface. YoRHa couldn't be around for longer than that. Androids in general, yeah they've been around for thousands of years.

Both are androids. The game doesn't really go in depth with their relationship, but according to the novels and side material they are each others most important person. The girl carrying the boy is actually not the main character, the boy is. After a certain event the boy turns into a crazy obsessive yandere.
