Middle Earth, followed closely by the Star Wars galaxy.

Hard to beat Middle Earth, but in junior high I was a total sucker for Pern.

Middle Earth, Warhammer 40k, and Azeroth(World of Warcraft).

For the last two while they may not be the most tight-knit on the lore, and in the case of WoW they retcon stuff whenever they like, I still enjoy the world that each game has created. There's a lot of neat stuff in them.

Tamriel (The Elder Scrolls). There's just so much to it I wouldn't even know where to begin.

Second place would probably go to Mass Effect. I love all the different species of the galaxy, and the different places. They did such a good job with the division between Citadel space and the Terminus systems, and of course places like the Perseus Veil.
