If they are original characters, sure. If there is a canon trans character and in the fic they are cis or if there is a canon cis character and in the fic they are trans, I think that needs to be tagged, just like if the character is genderbent. I wouldn't get upset at anyone not tagging otherwise, but I also wouldn't balk at a new category added with trans/trans, trans/cis, cis/cis, though maybe nonbinary and genderfluid combinations should be included too.
