I don't think Cas knew about the bigger picture up until 4.20, I think only the higher-up angels like Zachariah knew that the ultimate plan was to let Lucifer out, not keep him in. I think he did some digging around after 6.16 and found out what the real deal was, because Uriel had been fucking around behind his back, and then when he found out and tried to tell Dean about it they assreamed him.

Even before he rebelled at the end of season 4 I got the distinct impression that he wouldn't have gone along with a plot to let Lucifer free.
