I think the difference there is we know that a sort of afterlife does exist *in that universe*, so in that sense she's "singled out" this way.

That's sort of it for me. I am absolutely terrified of the idea of ceasing to exist if there really is no afterlife, but if that's the case, everyone's in the same boat. For Fred to live in a universe where life after death and a form of heaven is a reality but to be denied that and destroyed, through no real fault of her own, is highest-order nightmare fuel for me.

It also makes Angel and Spike's dilemma at the Deeper Well a lot grayer in retrospect (if you think her soul really was destroyed). Letting tens of thousands of people die to save one is obviously wrong, but if you're talking about the annihilation an eternal soul versus people who'd at least still exist metaphysically, how do you even put a value on that? Of course, neither Angel nor Spike knew about that at the time, but I'm sure it haunted them afterwards.
