As a side note, and I realize this is totally not the point of your post, AJ, Rarity, and a few other secondary characters have noticeably different accents. Rarity likes to speak in a vaguely "Canterlot" accent because she thinks it makes her sound sophisticated (think The Rain in Spain), but her sister and parents speak in the American Midwestern-ish accent that's typical of Ponyville. Interestingly, Twilight and her family don't have Canterlot accents, despite having presumably lived there their whole lives.

Applejack was already talked about - she speaks in more of an obvious Southern drawl, because this is a kids' show and she's a farmer.

One of the other two really noticeable differences is the Canterlot ponies, who speak in British accents (although I don't know enough about Britain to know which part). Again, it's because MLP is a kids' show and it's American, and Americans tend to connect that accent with being higher-class and with sophistication.

The other one is Zecora, who speaks in what I think is a Swahili accent...? But beyond the unfortunate implications of Zecora as a whole, I kind of love that her facial bone structure supports her accent: she's got a bigger sinus cavity than the other ponies, and her voice has a corresponding lush ring to it.

I actually really want to know where the "all farmers are southern" thing came from, because the American Breadbasket actually has straight-up perfect "broadcaster" accent. Seriously, like 90% of farm country sounds exactly like Walter Cronkite.

FYI, the accent of the Canterlot ponies is 'generic upper-class English'. Denotes class rather than region.
