Maybe it's more the OP's SPN OTP than the entire SPN fandom's OTP? Although next to Sam/Dean, it kind of is SPN's OTP.

It is SPN's OTP. Next to Wincest, D/C has more enthusiasm and a larger following behind it.

Honestly, from the perspective of someone who has never watched the show and has only heard about the fandom secondhand, it kind of looks like it is.

It isn't. Destiel is the fandom OTP.

Wincest used to be, but it's pretty passé now. A lot of the big name Wincest shippers left fandom and/or are inactive, and Castiel has gained tons of fans while it seems a lot of people have complaints about how Sam is being written.

If you see a fandom post about pairings on tumblr, Sherlock/John, Ten/Rose, and Dean/Cas are most likely to be mentioned from what I've seen.
