Sam didn't shot Madison in Supernatural 2.17, they faked her death, she escaped, and later met and teamed up with Heather Bram from Dresden Files 1.03 and they formed a "support-group" kind of thing. THey're both alive and well, and model-citizens.

The way the Grimm-abilities work and manifest had not been retconned at all with the second season. The thing still gets "activated" when another Grimm (preferably a family-member) dies or dying.
