I don't have hard and fast rules on that but in general if I'm shipping something that's crack or non canon they have to have some sort of interaction to base a relationship off of. I'm pretty all over the map and do what I want to do with shipping. In my experience most slash is based off of connections between the characters and often times that includes chemistry whether its supposed to be brotherly or not rarely matters. I can't really think of a slash ship where the characters have no chemistry or something to connect the two of them together to give people a reason to ship them.
