I haven't played it but here are my guesses

1. You can go on a date with at least one of them in game
2. They are vaguely humanoid and that is al it takes for some people

Those are all 4chan people though, not AO3 people

and skeleshipping definitely happens on AO3/tumblr parts of fandom

so there's gotta be something to it beyond 4chan deviancy

Honestly, for a lot of people, I think it's just the personalities and not the fact that they're skeletons. If they had just been human, or furry, or robots, etc., they'd still get their share of nsfw content. Both brothers can be woobified in some way, and that's a big factor in popularity for a lot of characters in similar circumstances (ex. Onceler, Loki, Snape, etc.).

Contrast the brothers with Mettaton EX, a character who is portrayed to be attractive in-universe, but doesn't receive as much fan attention as Sans does (about the same as Papyrus though). Compare with Asriel, another "woobie" character whose fan attention seems to be more from the furry community (more preference given to his "older" persona).

Yes, personalities is definitely a big factor in this. That said, I'm personally digging the look of a skeleton, so... Not anything from Undertale since I'm not familiar with it, but Discworld Death as depicted by Kidby and Lewis from Mystery Skulls Animated.

Magic glowing energy dicks. Or I suppose also magic glowing energy bajingo, or both! If that's your thing. I mean realistically skeletons shouldn't be able to stay together and move without muscles and tendons and cartilage, or talk without lungs and vocal cords and tongues.

So if you're going to suspend disbelief for walking talking skeletons, why draw the line at fucking?
