Okay, correct me if I'm wrong but I don't think there's a gender neutral word for sibling in Japanese? ("Kyoudai" means siblings OR brothers, that's the closest I can find.) And like I guess you could avoid "he" but I read that at least historically "kare" was originally gender neutral, and that's how they were using it?
I mean I think you're probably right the masculine language is there to make sure people don't think they're female, don't think that means it's there to make people think they're male. Also they're literally all sexless, which doesn't SEEM like a retcon thing, so I'm not really sure what them being "male" would even mean in that context.

You can choose to write it in kana or kanji, though. 兄弟 is 100% brothers, but きょうだい is a bit more neutral (and I've seen it used in a context where a JP fic writer was trying to dodge gendering a character). There's not really a neutral form of nii-san/nee-san/etc. to my knowledge but characters could simply refer to their siblings by name.

It's very easy to avoid using any pronouns at all in Japanese, though.

Neutral words for addressing siblings are a bit odd in Japanese, but the thing with the manga is that the characters don't actually see each other as siblings in any normal sense of the word, so they don't need to call each other sibling words. They just do because that's just what they call them to show that they respect them. But there are other gender neutral words for that the author could have chosen to have them use instead.
