I'm not sure I'm right, but I think the OP is trying to say that it wouldn't have made sense for Wincest to have been popular from the get go if the main characters hadn't been hot. Which is a dumb thing to say, since most fandoms have at least some portion of the fandom shipping the main characters, but at least has the backing that their relationship wasn't so played up as it was in later seasons.

There was a particular thing in the show about soulmates sharing a heaven, and it was pretty explicitly coded as a "usually happens to couples" thing that also happens to Sam and Dean, IIRC. So I assume they're referring to that.
