I got into two big fandoms recently and found myself perplexed at the main ships in both. Avengers was one. I can see where Steve/Tony is coming from, though it doesn't interest me personally, but Clint/Coulson? What? Is it something from the comics?

Not really. Clint had that cameo in Thor where he was just like "I don't know, I'm starting to root for this guy" to Coulson, and the ship exploded from there. It was big even before Avengers came out.

The two do interact in the comics, but until recently (like, February) it was minimal. Keep in mind that Coulson is actually a character created for MCU that they added to the comics later on, so all of his comic appearances are very recent. Also his comic characterization is... quite different.

They do actually have a bit more interaction in the 'Fury's Big Week' comic series that is a prequel to the Avengers movie. A little more interaction after the scene from the Thor movie (showing that it was Clint's idea to cut Thor loose after they captured him) and then there was a scene of the two of them sharing a car driving away from New Mexico, Clint driving Coulson napping. It doesn't seem like enough to launch a ship to me, but it's more than the single two lines over the comm in the movie.
