Yaoi = a genre of manga about romantic relationships between men. Notorious for clichéd plots and exaggerated seme/uke (super-manly top/super-feminine bottom) roles. Often has strange anatomy (http://wtfyaoianatomy.tumblr.com has plenty of examples, if you're interested).

Slash = Fanfiction that pairs two men together (or two women, but that's usually called femmeslash to differentiate it from slash with men). Sometimes uses common yaoi genre tropes, but not always (it depends on the writer). Referring to slash as "yaoi" is more likely to happen in anime & manga fandoms, although there has been some debate over whether that is appropriate. Referring to non-anime slash as "yaoi" will at best get you some strange looks and at worst start a flame war.
