I try to make my responses correspond in length to the comment I'm replying to. For instance, if someone just says, "This was great, loved it!" I'll probably just reply, "Thanks! :3" But if they tell me specific things they liked or quote back their favorite lines to me, I'll try for a more in-depth response, like what I was thinking when I wrote a particular bit they liked or my thoughts on how the characters behaved.

But that's just me. I don't think there are hard-and-fast social rules for this; do what feels right for you.
