It's both serious and sarcastic at the same time, ime? It's sort of like saying you're a Trekkie or a Moonie, you're saying you really really like something (sometimes to the point where you're willing to throw hundreds or thousands of dollars at it), but the very use of the term is conveying kind of a self-deprecating humor.

Waifus and Husbandos are kind of like being an OTT fan of a particular character. Some people are going to have Waifus/Husbandos and it consists of a minor crush on a fictional character, other people are going to own a love pillow of that character and buy them a birthday cake every year. It all depends on the person.
