Can you think of any examples of female characters who display traits that would be considered "macho" in men, either because that's just her personality/upbringing/culture, or as a defense mechanism?

To varying degrees: Faith from Buffy, Max from Dark Angel, Jo Harvelle from Supernatural. Though if you mean "COMPLETELY macho and not feminine at all," that's a bit more difficult. Vasquez, Ripley and Sarah Connor actually are the nearest examples that come to mind.
