It really, really, really depends on the specifics of the story.

If I can believe the justification (said justification can range from a well-developed story arc to a properly-specified AU to "the story's just that cracky and hilarious") for the change, I'll go for it.

If my reaction to the mischaracterization is "um...no? He's not like that. Why is he like that here?" and no answer is given, then I can't read it, period, no matter how well-written the story is otherwise.
