Getting into the Big Two can be...involved. It's easier now, with wikipedia and fandom communities and all, but I wouldn't blame someone for just not wanting to bother. If I may, some easy-to-get-into stories with breathing room and no outside reading required?

Thor: The Mighty Avenger -- it's two very small trades, 8 issues in total, that is completely separate from any other continuity. It stands alone completely as the story of Thor's origin and his romance with Jane. The writing is great, the art is lovely and makes me smile, and the characters are fun. The romance is pretty sweet and laid on in just the right doses.

Transmetropolitan--this isn't even remotely sweet, except for when it is. But it is a fantastic, fun, horrible, relevant story that...y'know, explaining it just doesn't work. It's strange and weird and about a terrible world inhabited by terrible people, and the protagonists is a terrible person, only he's driven by the need to tell the Truth on a grand scale, and there are actually really great female characters in this, which surprised me, but there they are. It pulls no punches and is related to nothing but itself, and it's a contained, finished story (maybe ten trades, maybe less?) that you can probably pick up from your library.

Bone -- another completed story, fantasy with funny animal elements. Widely considered a classic because it is. Features stupid stupid rate creatures.

Also: Fables, Anya's Ghost, Rapunzel's Revenge, and Hereville. Honorable mention goes to the current Hawkeye comic, which isn't completely a stand-alone, but can be read knowing nothing more than: "This guy's name is Hawkeye, his schtick is a bow and arrow, his day job is being a superhero."
