Except, no. Sex and gender are not the same thing and a person's gender can absolutely be different than their sex. Gender is not biological. She was just being a homophobic jerk while defending another homophobic jerk.

What's she protesting then? The person she's defending is a transphobic asshole who targeted and harassed a trans person at her work and was fired for that. If Rowling's words mean what you say they do, then they make no sense in terms of the person she is trying to defend. So no, I don't think she really means that, though she is putting that veneer on to try to keep looking good.

No, that woman was fired for tweeting that sex is real, binary and inmutable. She was fired for stating her beliefs. She never harrased nor misgendered anyone at work. She did misgender someone on Twitter, tought. Gregor Murray, a non-binary man suspended by the Commissioner for Ethical Standards in Public Life for repeatedly calling women cunts.

No, that was the judge's decision.

She was fired for tweeting. She did not harass anyone. The judge decided that because she tweeted these things that meant she was going to harass someone. That's some real pre-crime thought policing right there.

The specific question at issue in the arbitration was whether you have to treat people with respect in your workplace regardless of your personal beliefs wrt trans people. So the behavior she's defending is being able to disrespect and harass trans people in a workplace. Biological sex being immutable is not relevant or a defense on that question and citing it as a defense is pretty much just transphobic (or "massively, implausibly ignorant and also only choosing to listen to transphobes on the question for some reason")
