Here goes! I try to keep this as spoiler free as possible.
Four kids play a game. The game is more then simply a game, it bends time and space and enables you to save your world from the apocalypse, and make a new universe. It also turns out by playing the game you cause that apocalypse in the first place (not playing was never an option, time and space manipulation and all that).
These children end up in an unfortunate situation where the game is not only un-winnable, but one of the games baddies has ascended to become something that not only threatens their session, but multiple universes and the spaces inbetween. This effects not only their session, but the session that created theirs. The children are helped and hindered by the winners of the previous session, who wrote their names in our stars (the zodiac is effectively each of these players identification in their language) and offer both antagonism and essential help depending on their mood. Additional input is provided by both helpful creatures in the future who survived the mess, and a malevolent creature who serves a threat that might be even bigger then the game baddie. Also, there might be more then 4 humans involved in the game after all, as each kid discovers their respective caretaker has more to do with everything then they thought...
The cast is huge. Meta references within and to the author's other works abound. The narrative reads like you are playing a text based game, usually told from a constantly shifting second-person narrative perspective. The narrative deliberately jumps around in time, thought it all makes sense in the end if you read everything.
YOU HAVE TO READ EVERYTHING. Now, if you don't like this in the end, that's fine, it's not for you, oh well. But if you read it, and you don't read all of it (the intermissions are not really intermissions at all) you will be really fucking confused. You can't read a synopsis just to skip to where it is now, there's too much detail that is lost, and inconsequential facts become devastatingly important later. You really, really have to click on the "show pesterlog" parts. Most agree that to save yourself some time, if you are still reading at the page titled "WV: Ascend" and you aren't liking this yet, you should stop. But do read that far, this thing starts slow and picks up momentum as it goes along. This slow start is really necessary to hammer out some things you have to know. It stops slogging by that WV page I mentioned.
If you do end up trying again then stick around, enjoy the community. We have fanart. We have fan SONGS. we have so many fan songs there are now two official fan albums. And multiple official music albums. Homestuck is a true webcomic: it uses flash, it uses gifs, there are links to side things to read... and the music is fucking beautiful. You can hate homestuck and love the music. So much music there are over 16 albums now. You can listen to most of it for free.

I'll have to save it to try it out. The last thing I need is to get obsessed with something at the moment (it's not a good time). Also, I'm still wary of joining new fandoms. Still kinda recovering from the battlezone of nasty mean-ness that was the Harry Potter fandom.
But thank you for taking the time to tell me about this. Shows how devoted some of the fans are.

This isn't even the best flash. There are better ones, and that's in an objective sense: the author improves his craft. But it's a good taste to see that yes, while it starts slow, there's also stuff like this.

Homestuck fans argue in house a lot, usually over stupid stuff, but people who skip to act five are probably the ones who get the most disdain. They tend to ask the stupidest "wait, I'm confused" questions. Probably because they skipped 4 FREAKING ACTS.
Don't do that. Please. But really, the trolls are not the main characters, though in a story this long they have story arcs that have lasted longer then some webcomics are in length. Still, you have a lot of pages before they show up proper.
That said, they are some of the characters many fixate on. They are pretty freaking popular, and quite awesome. But if you read waiting for them to show up... it'd be a chore.

The trolls aren't the main characters? Weird. Almost everything I see about Homestuck involves the trolls. I just assumed they were the main characters, or frequently recurring side characters.

And...um there are lots of HS crossovers and lots of fans (like me) who like them. I suppose it depends on where you go...I've yet to see a storm of rabid fans yell at someone for making a crossover but then I'm not really that active and hardly ever go on Tumblr so

And I don't mind the occasional 'this is what I like and why I like it' but the constant 'Homestuck! Homestuck! I will now talk about it at great length and also compare everything you ever say to Homestuck!' is what made me hate it.
